Archaeological sites carry an array of aDNA data that, if properly used, can contribute to our understanding of early farming. In this study, shotgun sequencing and mapping of ancient DNA was used to recover a section of ancient human mitochondrial DNA (mtDNA) from the human remains recovered from the Middle Neolithic site of Talheim, Germany. We combined this evidence with mtDNA and Y-chromosome (Y-chromosome) sequences recovered from later Neolithic sites in Europe to gain insights into the spread of farming into Europe in a time frame when there were no written records to document the spread of technology and knowledge.
During the past decade, aDNA has been increasingly studied. However, there are still few studies that are based on ancient DNA derived from the remains of vertebrate animals. These studies are fundamental to the study of paleontology, archaeological culture, as well as to the study of human evolution. The purpose of this article is to elucidate the methodology of obtaining aDNA and to summarize its potential for paleontological and archaeological research. The material used for the study was a teeth sample from a cave bear (Ursus arctos) from the Pleistocene of the Korean Peninsula. In this article, we provide information on the process of extracting DNA, the process of sequencing, and the validation of the amplified fragments. After the validation, we analyzed the genetic composition of the cave bear sample by extracting DNA from one of its teeth. This article is also the first to report in detail the process of aDNA extraction from archaeological samples.
Plants and animals are the most obvious sources of DNA in ancient DNA research, as they make up more than 90% of the total DNA in a specimen. Plant and animal remains from archaeological sites provide a wide range of applications in archaeological science. Archaeologists can use these remains to 1) determine how particular species were used in the past, 2) assess the size and diversity of populations, 3) investigate cultural heritage, 4) assist in the reconstruction of past environments, 5) study the relationships between and among humans, and 6) track the migration of populations. Ancient plant DNA (aDNA) can be directly used as evidence to suggest that an association between plants and people existed in the past. In addition, plant remains can also be used as evidence of trade between cultures or geographic regions. A range of aDNA studies on plants and animals are summarized, using examples from the published literature to illustrate the diverse aspects of ancient DNA studies with plant and animal remains.
